大白鼠离体器官冻融组织学观察及理化特性

摘  要:用AC15型复射式检流计和Olympus显微镜组织学观察研究以及图像分析了不同的温度浸冻大白鼠(Wistar)在体尾和离体心、肺、肝、脾、肾、脑的组织温度和组织有无显著变化。结果离体器官的组织温度和组织有显著变化。组织过冷点温度(-4.4±0.69℃~-12.13±1.87℃)与鼠尾的组织过冷点相比有统计学意义(p<0.01)。从过冷点到组织冻结点均为-0.60℃,时间均为2秒,我们认为细胞、组织、器官复温速率为120~360℃/min为宜。冷液浸冻组织以-12℃~-20℃为宜,否则组织将不出现过冷现象,或者细胞、组织变性,处于亚稳定状态。AC-15 thermocuple, tharmometer, olympus microscope and picture analyse were used to study the changes in temperature and histology structure of tail in vivo and isolated heart, lung, liver spleen, kidney and brain of Wistar rats after freezing at different temperatures the results showed that: ①There was marked crrand in temperature and remarkable change in histology structure between tail in vivo and isolated heart lung, liver, kidney and brain. ②)The freezing point was -0.60℃ and the time from supercooling point to freezing point was 2 seconds. ③These results suggested that: temperature-recovering rate of cell, tissue and organ should be at 2° to 6°/sec and the preparation temperature of cold solution freezing tissue should be at - 12℃ to - 20℃ other- wise the tissue would not show supecoolng and the cell tissue would show denaturation and become in a substable state.
