摘  要:目的:研究盐酸小檗碱抗假丝酵母菌及抗肿瘤活性,为临床用药提供依据。方法:依据NCCLS的M27-A2标准方案测定标准株对盐酸小檗碱的药物敏感性;MTT法测定盐酸小檗碱对细胞生长的抑制作用;流式细胞仪检测盐酸小檗碱对人舌癌细胞Tca8113凋亡率的影响。结果:盐酸小檗碱具有抗假丝酵母菌活性,能抑制Tca8113细胞生长;浓度为160、80、40、20、10μg/ml的盐酸小檗碱作用于Tca8113细胞48h后凋亡率分别为32.3%、24.6%、13.3%、6.35%、6.51%,对照为8.46%。结论:盐酸小檗碱(C20H18ClNO4)体外具有抗假丝酵母菌活性,且能抑制舌癌细胞Tca8113的生长,诱导舌癌细胞Tca8113的凋亡。Objective :To study antifungal activities of hydrochloric berherine( C20H18 ClNO4 ) against oral Candida spp by NCCLS M27-A2 method and to study the growth inhibition and apoptosis induction effects of hydrochloric berberine on human tonge carcinoma cells Tca8113 in vitro. Methods:The in vitro susceptibilities of Candida spp to fluconazole and hydrochloric berherine were determined by NCCLS M27-A2 method. Tca8113 cell proliferation ability was measured by MTT assay. Apoptotic rate of the cells was analyzed by flow cytometry. Results: Hydrochloric berberine exhibited antifungal activity. MTT assay results showed that hydrochloric berberine inhibited Tca8113 cells growth. After treated with 160, 80, 40, 20, 10 μg,/ml hydrochloric berherine for48 h, FCM assay revealed that the apoptotie rate were 32.3%, 24.6% , 13.3%, 6.35%, 6.51% respectively. Conclusion: Hydrochloric berberine might be used for the prevention and treatment of oral eandidiasis. Hydrochloric berberine has inhibitory effects on the growth of Tca8113 and induces the apoptosis of human tonge carcinoma cells Tca8113.
