摘  要:目的:探讨高浓度同型半胱氨酸(homocysteine,Hcy)对人脐静脉内皮细胞(HUVEC)CRL-1730一氧化氮合酶(eNOS)mRNA表达的影响。方法:用不同浓度Hcy(0~5.0mmol/L)刺激HUVEC CRL-1730,分别作用1~24h,收集细胞,提取总RNA,逆转录得到cDNA,PCR扩增eNOS基因片段,2%琼脂糖凝胶电泳分离PCR产物,计算相对含量。结果:随着Hcy浓度增高HUVEC CRL-1730eNOS mRNA表达逐渐降低;随着Hcy作用时间的延长,HUVEC CRL-1730eNOS mRNA表达降低。病理浓度(0.1mmol/L)及毒性药理浓度(5.0mmol/L)Hcy与生理浓度Hcy(0.01mmol/L)相比可显著降低HUVEC CRL-1730eNOS mRNA的表达。结论:高浓度Hcy可能通过降低eNOS水平,减少NO合成,导致血管内皮功能损伤。Objective: To investigate the effect of homocysteine (Hcy) at different concentrations on the eNOS mRNA in cultured human umbilical vein endothelial cells (HUVEC), CRL-1730. Methods:The HUVEC CRL-1730 was cultured with different concentrations of Hcy (0 to 5.0 mmol/L)for 1 to 24 hours, and then the cells were collected to isolate the total RNA. RT-PCR and 2% agarose gel electrophoresis were performed to get the eNOS fragments. The results were calculated. Results: The eNOS mRNA was significantly depressed with gradually increased concentration of Hcy. Hcy significantly depressed the eNOS mRNA in pathological concentration (0.1 mmol/L) and toxic concentration(5.0 mmol/L) compared with the physiological concentration (0.01 mmol/L). Conclusion:Hcy in higher concentration may lead to the endothelial dysfunction by depressing eNOS and reducing the production of NO.
