摘 要:目的研究γ射线对人外周血淋巴细胞cx43和ANLN基因转录表达的影响。方法对数生长期的淋巴细胞,分别给予1、2、3、4、5、6 Gy的^60Coγ射线照射,照射后12h,以及2Gy照射后4、8、12、24、36、48、72h,分别提取总RNA,反转录成cDNA。利用实时荧光定量PCR技术,检测各组cx43和ANLN基因表达改变。结果人外周血淋巴细胞cx43 mRNA表达水平在2Gy照射后4、8、12h明显增高,分别为对照组(未照射组)的6.74、9.06、7.22倍(P〈0.05);24~72h,其表达水平与对照组相比没有明显变化。1、2、3、4、5、6Gy剂量照射后12h,cx43 mRNA表达水平显著增高(P〈0.05)。ANLN mRoNA表达水平在2Gy不同时间点及1~5Gy照射后12h,表达降低(P〈0.05),6Gy照射后12h其表达开始升高,为对照组的6.08倍(P〈0.05)。结论γ射线照射2Gy不同时间点及不同剂量照射后12h,cx43基因表达上调,ANLN基因表达下调。1~3Gy剂量照射后12h,cx43 mRNA表达在此范围内有时间和剂量的依赖性。cx43可能会发展为核事故受照射人员的分子生物学剂量标记物。Objective To study the effect of γ-rays on transcription of cx43 and ANLN gene in human peripheral blood lymph ceils. Methods Lymphocytes in logarithmic growing phase were irradiated under 1, 2, 3, 4, 5, and 6 Gy of γ-rays, respectively. Total RNA was extracted before irradiation and 12 h after irradiation at each irradiation dose above, and 4, 8, 12, 24, 36, 48, and 72 h after irradiation at 2 Gy dose as well. Extracted RNA was reversed into cDNA by reverse transcription. Fluorescent quantitative PCR was used to detect the expression of cx43 and ANLN taRo NA. Results Compared with control, at the dose of 2 Gy, the level of cx43 mRNA in lymphocytes was increased significantly after 4, 8, 12 h exposure, up to 6. 74, 9.06, 7.22 fold respectively (P 〈 0. 05), whereas there appeared no significant changes in the time course of 24-72 h. Twelve hours after irradiation at the dose range of 1, 2, 3, 4, 5, and 6 Gy, the level of cx43 mRNA expression was significantly increased ( P 〈 0. 05 ) . The level of ANLN mRNA expression was decreased both at the dose of 2 Gy after different time points and at the dose range of 1-5 Gy after 12 h (P 〈 0. 05) . However, after 12 h irradiation at the dose of 6 Gy, the expression level of ANLN mRNA was increased up to 6.08 fold (P 〈 0. 05 ). Conclusion At 2 Gy dose of γ-rays with different time courses, or after 12 h irradiation under different dose ranges, expression of cx43 mRNA is upregulated, whereas ANLN mRNA is downregulated. 12 h irradiation at the dose range of 1-3 Gy, expression of cx43 mRNA presents a dose and time-dependant manner. Cx43 may become into a molecular biological dose biomarker for people who are subject to radiation due to nuclear accident. To this end further intensive investigation need to be carried out.
