摘 要:建立了一种反相高效液相色谱法同时分离和测定金银花和金银花露中绿原酸和4种黄酮类化合物的含量.优化样品提取条件和实验条件下,采用甲醇-水(H3PO4调pH=2.6)梯度洗脱,外标法定量.结果显示绿原酸、槲皮素、金丝桃苷、木犀草素、芹菜素的线性范围分别为0.1~50,0.055~100,0.05~200,0.05~100,0.1~100mg/L(R>0.999),回收率分别为94.40%,100.72%,97.60%和96.80%(相对标准偏差小于1.2%).此实验方法简便,快速,线性关系良好,可作为金银花品质的测定.A method to simultaneously separate and determinate chlorogenic acid and four flavones was established based on reversed-phase high performance liquid chromatography. Under the optimized and experimental conditions, a mixture of MeOH-water, with pH adjusted to 2.6 with H3PO4, was used as the mobile phase, and external standard was also employed The results showed the linearity range was 0. 1-50, 0. 055-100, 0.05-200, 0.05-100 and 0.1-100 g/mL, respectively, for chlorogenic acid, Quercetin, Hyperoside, Luteocin and Quercetin (R〉0.999) and their recoveries were 94.40%, 100.72%, 97.60% and 96.80% (RSD〈1. 2%). This method proved to be sensitive, accurate, rapid and reproducible. It could be applied to determine the quality of Flos lonicerae.
