摘  要:目的观察人胚胎生殖细胞移植于大鼠急性心肌梗死区后的存活率和增殖分化能力,探讨人胚胎生殖细胞异种异体移植的可行性。方法取5~9周人胚胎生殖脊细胞,分离、体外培养获得胚胎生殖细胞;用SD大鼠建立心肌梗死模型,分为对照组、移植组。分别在移植后1 d和1、2、4周处死大鼠,以抗人细胞核抗体MAB1281作为示踪剂,观察转录因子Nkx-2.5在移植大鼠心脏组织的阳性表达;分析人胚胎生殖细胞在宿主体内转化为心肌细胞的能力。结果成功建立大鼠心肌梗死模型;移植组心肌梗死区抗人细胞核抗体MAB1281和转录因子Nkx-2.5均呈阳性表达,对照组均为阴性表达。结论人胚胎生殖细胞植入心肌组织后,不仅在心肌内存活而且表现出向心肌细胞分化的特性。Objective To study the possibility of xenogeneic transplantation of human embryonic germ(EG) cells by observing the surviving rate and transforming abilities of transplanted human EG cells through different transplantion methods.Methods The gonadal ridges and dorsal mesenteries from human embryos aged 5~9 weeks were taken to serve as transplanted cells.A model of myocardial infarct in the rat was created.It’s divided into 2 groups:The transplanted group and control group.These rats were killed respectively 1 day and 1,2,4 weeks after transplantation.Anti-human nuclei MAB1281 was used as tracer agent to identify the distribution of transplanted cells and the positive pre-sentation of transcription factors Nkx-2.5 was observed.Results In the transplanted group,human cel-lular nuclei MAB1281 and transcription factor Nkx-2.5 in myocardial infarction area showed positive presentation.Conclusion Human EG cells transplanted into rats’ myocardium can live and differenti-ate into cardiomyocytes.This experiment indicates that it is feasible to xenogeneically transplant human EG cells into rats’ myocardium.
