摘 要:目的:研究红霉素对小鼠海马神经元电压依赖性钾电流的影响。方法:采用分离新生小鼠海马细胞和全细胞膜片钳技术的方法,观察给予红霉素前后海马神经元细胞电压依赖性钾电流的变化和电流-电压曲线的改变。结果:红霉素可抑制小鼠海马细胞电压依赖性钾电流,使电流峰值由(810.67±250.86)pA下降至(529.96±171.83)pA(P<0.05);给予系列去极化脉冲刺激海马神经元,随着刺激电压强度的增强,钾外向电流增加,电压-电流曲线右移,但不改变曲线的形态。结论:红霉素可抑制小鼠海马神经元电压依赖性钾电流,并可能通过中枢机制来影响其调节胃肠运动的效应。OBJECTIVE: To study the effect of erythromycin on the voltage- dependent K ^+ current in hippocampal neurons of mice. METHODS: Isolation of mice' s hippocammal neurons cells and whole - cell patch - clamp techniques were applied to study the change of voltage - dependent K^ + current of hippocampal neurons and the current voltage curves in mice before and after treatment with erythromycin. RESULTS: Erythromycin inhibited the voltage - dependent K^ + current of mice's hippocampal neurons, and it down regulated the peak current from (810.67 ±250.86) pA to (529.96 ±171.83) pA (P 〈 0.05) . During a series of depolarized pulse stimulation on hippocampal neurons, the K ^+ outward current increased with the increase of the intensity of the stimulation voltage, and the voltage - current curve shifted right, but the shape of the curve remained unchanged. CONCLUSIONS: Erythromycin can inhibit the voltagedependent K ^+ current of hippocampal neurons in mice and regulatory effect on the motility of gastrointestinal tract is influenced by the mechanism of central nervous system.
