摘  要:目的通过在视网膜组织三维立体培养系统中加入不同浓度的川芎嗪溶液,观察川芎嗪对视网膜神经节细胞轴突再生的作用。方法建立体外培养的大鼠视网膜组织三维立体培养系统,将1~3d新生远交群SD(Sprague Dawley)大鼠视网膜切成0.5mm×0.5mm大小的视网膜组织,加入不同浓度(0.125、0.25、0.5、1.0g/L)川芎嗪溶液后,在相差显微镜下动态观察视网膜神经节细胞轴突的生长情况,于加药后第3、6和9天记录再生轴突的数目及长度。结果与对照组相比,各浓度川芎嗪对视网膜神经节细胞轴突生长均有促进作用,以0.5g/L浓度效果最明显,差异有统计学意义(P<0.05)。免疫组织化学染色显示视网膜神经节细胞的轴突具有明显再生现象。结论一定剂量范围的川芎嗪可促进视网膜神经节细胞轴突再生和伸长。Objective To investigate the eflect of tetramethylpyrazine on the axon regeneration of retinal ganglion cells(RGCs) in vitro through adding different concentrations of tetramethylpyrazine to the three-dimensional culture of rat's retinal tissue system. Methods The retinal tissues of Sprague Dawley rats aged from 1 to 3 days were cut into pieces of 0.5 mm×0.5 mm, and cultured in a three-dimensional culture system. The effect of tetramethylpyrazine (0. 125, 0.25, 0.5, 1.0 g/L) on the growing of RGC axons was investigated under the fluorescence inverted microscope with immunohistochemieal staining. The number and length of outgrowing RGC axons was observed at day 3,6 and 9 respectively. Results The number and length of regrowing axons to all of the tetramethylpyrazine treated groups was all larger than that in the control group and the effect to 0.5 g/L tetramethylpyrazine was :the most obvious ( P 〈 0.05 ). lmmunochemistic positive fibers showed to be regenerated axons. Conclusions Tetramethylpyrazine improved the regeneration and enlongation of RGC axons in the three-dimensional culture system at certain concentrations.
