摘  要:利用碱性蛋白酶酶解技术对新鲜猪血进行血红素提取。采用响应面分析法对酶解时间、酶解温度、酶解pH、酶与底物浓度比等因素进行了优化。结果表明,酶解最佳条件为:酶解温度59.3℃、酶解pH8.6和酶解时间125.0 min和酶底物浓度比4 680.0 U,在此条件下,血红素的得率为72%、提取粗品纯度为33.8%。Alkaline protease enzyme technology was used to carry out fresh pig's blood hemoglobin extraction. Response Surface Methodology was used to optimize the factors which were digestion,digestion temperature, digestion pH,enzyme and substrate concentration and other factors. The results showed that the best conditions for the digestion were as followed: digestion temperature was 59.3℃, pH was 8.6, hydrolysis enzymatic time was 125.0 minutes and concentration than the end 4 680.0U. Under these conditions, the hemoglobin rate was 72%, the purity of crude extract reached to .33.8 %.
