摘 要:目的探讨阿霉素肾硬化大鼠转化生长因子-β1(TGF-β1)表达和细胞凋亡的相关性,及商陆对其的影响。方法采用单侧肾切除并阿霉素尾静脉注射的方法构建肾小球硬化大鼠模型。36只SD大鼠随机分为正常对照组,模型组,商陆治疗组,分别给予灌胃治疗,连续93d。观察肾小球病理改变,采用免疫组化法检测各组大鼠肾小球TGF-β1的表达;TUNEL法检测肾小球细胞凋亡,并分析TGF-β1水平和细胞凋亡的相关性。结果与模型组比较,商陆水煎剂能明显改善肾小球病理损害程度,降低肾小球硬化指数(90.15±19.73对40.49±8.86);显著降低阿霉素肾硬化大鼠肾小球TGF-β1的表达(6.34±0.44对2.76±0.73,P<0.01)及肾小球细胞凋亡指数(63.62±9.92对34.46±7.19,P<0.01);并且肾小球细胞凋亡指数和TGF-β1的表达(r=0.658,P<0.01)、肾小球硬化指数(r=0.768,P<0.01)间均存在相关性。结论(1)阿霉素肾小球硬化大鼠肾小球TGF-β1的表达与肾小球实质细胞凋亡间存在显著相关性。(2)商陆在降低肾小球TGF-β1的表达的同时降低了肾小球细胞过度凋亡,改善了肾小球病理损害,保护了肾功能。Objective To explore the correlation between the expression of transforming growth factor - β1 ( TGF - β1) and apoptosis, and the effects of Phytolaccae on TGF - β1 expression and apoptosis in rats with Adriamycin - induced glomerulosclerosis. Methods Forty - eight male SD rats were randomly divided into three groups: control group, model group and Phytolaccae - treated groups. The Glomerulosclerosis rats model was established via uninephrectomy and were injected intravenously with Adriamycin 5 rag/( kg · d), 3 mg/( kg · d) separately on the 7th and 28th day after uninephrectomy respectively. On the subsequent days, the rats were treated with gastric - perfusion respectively for 93 days. At the end of the treatment, renal tissue pathology were analyzed. The TGF - β1 expression was detected by immunohistochemistry. Apoptosis was detected by tunel method. Meanwhile, glomerulosclerosis index and the apoptosis index were calculated. Results Compared to the model group rats, the animals of Phytolaccae - treated groups showed a significant decrease in glomerulosclerosis index(90.15 ± 19.73 vs 40.49 ±8.86)and the apoptosis index(63.62±9.92 vs 34.46±7.19 ,P 〈0.01 ) and had a statistically significant increase in expression of TGF -β1 (6.34 ± 0. 44 vs 2.76 ±0.73, P 〈 0.01 ). The apoptosis index correlated with TGF - β1 expression ( r = 0. 658, P 〈 0.01 ) and the glomerulosclerosis index( r = 0. 768, P 〈 0.01 ). Conclusions There is a positive correlation between TGF - β1 expression or the glomerulosclerosis index and renal apoptotic cells. Phytolaccae can significantly decrease renal decrease, renal tissue morphology damage, TGF - β1 expression , over -apoptotic and ameliorate glomerulosclerosis of adriamycin - induced Glomerulosclerosis.
