摘 要:目的研究哮喘发作患者血浆和豚鼠哮喘模型血浆、肺组织内皮素1(ET1)、肿瘤坏死因子α(TNFα)、丙二醛(MDA)和超氧化物歧化酶(SOD)的变化及一氧化氮(NO)合成酶抑制剂亚硝基左旋精胺酸甲酯(LNAME)对其影响。方法选择患者及建立豚鼠哮喘模型,应用放射免疫法测定ET1,酶联法测定TNFα,硫代巴比妥酸法及羟胺氧化法测MDA和SOD。结果哮喘患者血浆ET1、TNFα及MDA分别为33±15(×109g/L),3.18±0.65(×109g/L),8.2±1.5(×106mol/L)显著高于对照组14±4(×109g/L),2.34±0.08(×109g/L),4.9±1.3(×106mol/L,P<0.01);而哮喘患者血浆SOD活力为135±26(×10-3NU/L),显著低于对照组161±19(×10-3NU/L,P<0.01)。豚鼠模型血浆、肺组织ET1含量为127±8(×109g/L),243±58(×108g/g);TNFα含量为3.50±0.13(×109g/L),32.4±3.5(×108g/g);MDA含量为213±43(×106mol/L?Objective To investigate the role of the ET 1、TNF α and oxidative radicals of MDA and SOD in the pathogenesis of asthma and the effect of the nitric oxide synthase inhibitor, L NAME to above factors. Method ET 1 was detected by the radio imunoassay and TNF α was detected by ELISA, SOD and MDA were detected by using thiobarbituric acid method. Result The contents of ET 1, TNF( andMDA in the serum of the asthmatic patients and in the serum and lung tissue of asthma models of guinea pigs were significantly increased, in the contrast ,the content of SOD was decreased compared with those in the controls( P <0.01). All the ehanges in the serum and in the lung tissues of the asthma models of guinea pigs returned to the levels of the controls by dexamethasone inhalation or L NAME by intraperitoneal injection. Conclusion ET 1, TNFα and the oxidative radicals and NO may play important roles in the pathogenesis of asthma, L NAME, which is similar to the dexathamethsone, may either decrease the content of NO or decrease the content of ET 1, TNFα and oxidative radicals in asthma.
