摘 要:分别以5龄幼虫的精巢和卵巢构建了2个家蚕cDNA文库。利用表达序列标签方法分析参与家蚕性腺发育的基因,通过测序获得16 737条ESTs(精巢8 407条,卵巢8 330条),拼接这些ESTs共得到2 107个重叠群(con-tig)和3 532个单拷贝(singlet)。将这些转录物与GenBank非冗余蛋白质库比较,结果显示约34.3%的基因与数据库中蛋白质具有相似性。功能注释表明蛋白质合成相关基因、精巢特异基因等在文库中大量存在。基因本体(gene ontology)功能分类显示精巢与卵巢基因表达谱差异较大。研究结果为理解家蚕性腺发育提供了信息。Two silkworm cDNA (complementary DNA) libraries were constructed from testes and ovaries of the silkworm Bombyx mori, respectively. An expressed sequence tag (EST) analysis was performed to identify major genes involved in silkworm gonad development. 16 737 ESTs were generated by sequencing cDNA clones from the two libraries (8 407 from testis library and 8 330 from ovary library), and assembled into 2 107 contigs and 3 532 singlets. As compared to the non-redundant protein database in GenBank, approximately 34.3% of the transcripts matched proteins in the database. Functional annotations showed that genes related to protein synthesis and testis-specific proteins were enriched in both libraries. Gene ontology classification indicated that there was a remarkable difference in gene expression profiles between testes and ovaries. These results provide useful fundamental information for further studies on silkworm gonad development.
