摘  要:采用实时定量RT-PCR方法,对丝素轻链基因(fib-L)、丝素重链基因(fib-H)、P25基因在家蚕幼虫不同组织和不同发育时期的表达进行了定量分析。结果发现3种丝素基因除主要在5龄幼虫的后部丝腺中表达外,在其它组织如脂肪体和中部丝腺中也有一定程度的表达;在4龄眠期的后部丝腺中3种丝素基因也有一定的表达水平,其中fib-L在这一时期的表达量较高。同时发现在5龄第3天和第5天的后部丝腺中,fib-H、fib-L与P25在mR-NA水平上的摩尔比分别为9∶18∶1和19∶32∶1,推测丝素基因的表达可能具有转录后调控,此结果说明家蚕丝素基因的表达可能具有更加精细的调控机制。In this paper, the expression of fibroin heavy chain gene (fib-H) ,fibroin light chain gene (fib-L) and P25 gene in different tissues and different developmental stages was investigated at the mRNA level using quantitative real-time RT-PCR method. The results indicated that the expression of three fibroin genes was very high in the posterior silk gland during the fifth larval stage. Except for the posterior silk gland, there was a cer- tain degree of expression in other tissues, such as fat body and middle silk gland. At the fourth molting stage, there also existed the expression of fibroin genes, of which the expression of L-chain gene was much higher than that of the other two fibroin genes. At the same time, we found that molar ratios of the three fibroin genes' expression in the posterior silk gland at day 3 and day 5 of fifth instar were 9,18:1 and 19.32: 1 respectively at the level of mRNA. These results indicate that the regulatory mechanism on expression of fibroin genes is more delicate than what we ever knew.
