摘 要:【目的】建立加味芍药甘草颗粒中芍药苷及甘草酸的含量测定方法。【方法】采用反相高效液相色谱法,色谱条件:乙腈(A)-10 g/L冰醋酸水溶液(B)(调pH3.5),流速1.0 mL/min;检测波长250 nm,柱温为室温,进样量20μL。梯度洗脱程序:体积分数18%A、0~5 min,体积分数19%~29%A、6~10 min,体积分数30%~45%A、11~25 min(A+B=100%)。【结果】芍药苷及甘草酸分别在1.916~122.6、2.625~168μg/mL范围内成良好线性关系,平均回收率芍药苷为101.12%,甘草酸为99.68%。【结论】本测定方法简便可行、重复性好,可用于加味芍药甘草颗粒中有效成分芍药苷和甘草酸的含量测定。Objective To establish a method for the determination of peoniflorin and glycyrrhizic acid in modified Shaoyao Gancao Granules.Methods Reversed-phase high performance liquid chromatography(HPLC) was used.The chromatographic condition was as follows: Diamonsil C18 column(4.6mm ×250mm,5μm),column temperature being room temperature,mobile phase consisting of acetonitrile(A)-10g/L acetic acid(B) with adjusting pH value being 3.5,flowing rate at 1.0mL/min,detective wavelength at 250nm,and the injection volume being 20μL.The gradient elution procedure was as follows: elution with 18% A for 0~5 min,elution with 19%~29% A for 6~10 min,elution with 30%~45% A for 11~25min,and A+B=100%.Results Peoniflorin at the concentration of 1.916~122.6μg/L and glycyrrhizic acid at the concentration of 2.625~168μg/L showed a good linearity.The mean recovery was 101.12% for peoniflorin and 99.68% for glycyrrhizic acid.Conclusion This method is simple and reproducible,and can be used for the determination of peoniflorin and glycyrrhizic acid in modified Shaoyao Gancao Granules.
