摘　　要：【目的】建立食蟹猴糖尿病模型,并确定诊断标准。【方法】选用雄性食蟹猴8只,随机分为正常对照组和模型组;采用一次性静脉注射链脲佐菌素(STZ,剂量为45 mg/kg)法造模,正常对照组一次性静脉注射柠檬酸缓冲液(剂量为45mg/kg);观察造模前后各组猴的摄食量、饮水量、尿量、体质量变化,检测各组尿糖、空腹血糖、糖耐量、甘油三酯、总胆固醇、C肽、谷氨酸脱羧酶抗体(GAD-Ab)的变化,并进行胰腺组织病理学检查。【结果】造模后模型组摄食量、饮水量及尿量显著增加,体质量、C肽含量显著下降(均P<0.01),甘油三酯和总胆固醇均无显著变化(P>0.05);造模后第2周空腹血糖升高,尿糖由阴性变为"++++";第5周出现糖耐量异常,第7周空腹血糖与正常组比较有显著性差异(P<0.01),GAD-Ab阳性;胰腺组织病理检查结果显示模型组胰岛萎缩、纤维化,而正常对照组胰腺组织无病理改变。【结论】成功建立Ⅰ型糖尿病食蟹猴模型。Objective To explore a method for establishing the cynomolgus monkey diabetic models and to set up its diagnostic criteria.Methods Eight male cynomolgus monkeys were randomized into the normal group and the model group.The cynomolgus monkey diabetic models were induced by one-dose intravenous injection of streptozocin（STZ,at the dose of 45mg/kg）.The normal control group was given citrate buffer（at the dose of 45mg/kg）.Before and after modeling,the changes of food intake volume,water intake volume,urine volume and body weight were observed.Meanwhile,the changes of urine glucose,fasting glucose,glucose tolerance,triglyceride,total cholesterol,C-peptide and glutamic acid decarboxylase antibody（GAD-Ab） were examined,and the pathological change of pancreas was checked.Results In the model group after modeling,the food intake volume,water intake volume and urine volume were increased,and body weight as well as C-peptide content was decreased （P〈0.01 compared with the normal control group）.There were no obvious changes of triglyceride and total cholesterol（P〉0.05）.Two weeks after the modeling,fasting glucose was increased,negative urine glucose become strong positive（＋＋＋＋）; 5 weeks after the modeling,glucose tolerance became abnormal;7 weeks after the modeling,fasting glucose in the model group differed from that in the normal group（P〈0.01）,GAD-Ab became positive,and the pancreatic pathological results showed that atrophy and fibrosis were found in the pancreas of the model group,but the pancreas in the normal group remained normal.Conclusion The cynomolgus monkey diabetic model has been established successfully.
