摘 要:目的研究白细胞介素-9(IL-9)和白细胞介素-5(IL-5)在支气管哮喘气道炎症中的作用。方法收集支气管哮喘患者痰液40例,正常对照组20例。用夹心酶联免疫吸附法测定诱导痰上清液IL-9、IL-5及IL-8浓度。结果哮喘发作期组IL-9、嗜酸性粒细胞(EOS)、IL-5明显高于哮喘缓解期组(P<0.01);哮喘发作期组和缓解期组的IL-9、IL-5、EOS明显高于正常对照组(P<0.01)。哮喘发作期组IL-9浓度分别与EOS和IL-5浓度呈正相关(r=0.39,P=0.025;r=0.542,P=0.001),哮喘发作期组IL-8浓度与中性粒细胞百分比呈正相关(r=0.881, P<0.001);在哮喘发作期的中、重度患者中,IL-9与IIL-8水平呈正相关(r=0.528,P=0.002)。结论IL-9与哮喘气道内的EOS、IL-5密切相关,在哮喘发作期尤为明显,表明IL-9和IL-5参与哮喘患者气道炎症过程。Objective To investigate the role of interleukin-9 (,IL-9) and interleukin-5 (IL-5) in the pathogenesis of airway inflammation in asthma. Methods Induced sputum was obtained from 40 asthmatics. 20 healthy individuals were enrolled as controls. IL-9, IL-5 and IL-8 in sputum supernatants were assayed by sandwich enzyme-linked immunosorbent assay(ELISA). Results The counts of EOS,the levels of IL-9,IL-5 in induced sputum of the asthmatics were significantly higher than those in the controls(P 〈 0.01 ). Furthermore, these parameters were higher in the asthmatics of acute exacerbation than those of stable phase (P 〈 0. 01 ). In the acute exacerbated asthmatics,a positive correlation was found between the level of IL-9 and the percentage of eosinophile ( r = 0.39, P = 0. 025 ) , the level of IL-5 and IL-9 ( r = 0. 542, P = 0. 001 ) ,the level of IL-8 and the percentage of neutrophil (r =0. 881 ,P 〈0. 001 ) ,and the level of IL-9 and IL-8 ( r = 0. 528, P = 0. 002). Conclusion IL-9 is increased in the induced sputum from asthmatics, especially in exacerbation with asthma, suggesting that IL-9 and IL-5 may participate in the modulation of asthmatic airway inflammation.
