摘  要:目的建立中药HPLC指纹图谱在线专家系统分离系统,在该系统的指导下建立田基黄药材指纹图谱方法,验证专家系统的正确性。方法色谱柱:依利特Hypersil ODS2柱(250mm×4.6mm,5μm),流动相:乙腈-H3PO4水溶液(pH3.O),线性梯度洗脱,检测波长:275nm,柱温:35℃,流速:1.0mL·min^-1,分析时间:60min。结果由专家系统指导得到的色谱条件较理想,证明了系统的正确性。田基黄指纹图谱有16个共有峰。用药典委颁布的指纹图谱相似度计算软件,以夹角余弦为测度,测得20批田基黄药材指纹图谱相似度绝大部分在0.90~1.00之间。结论建立的田基黄药材HPLC指纹图谱分析方法准确、简便、重复性好,初步验证了专家系统的正确性。Objective To establish the separation system of Expert System of Traditional Chinese Medicine HPLC Fingerprint online, to establish chromatographic fingerprint method of Hypericum japonicwn and to prove the correctness of the Expert System. Methods Separation was performed on a Hypersil ODS2 (250 mm × 4. 6 mm, 5 μm) analytical column with mobile phase consisting of acetonitrile-buffer mintion (water-KH2PO4-H3PO4 , pH 3.0) with linear gradient elution. Detec- tion wavelength: 275 nm; column temperature: 35 ℃ ; flow rate.. 1.0 mL·min^-1 ; and analysis time: 60 min. Results The chromatographic condition obtained under the direction of Expert System was ideal, which proved the correctness of the Expert System. Sixteen co-peaks on the HPI.C fingerprints of Hypericum japonicurn were obtained. The similarities were determined by cosine. Most similarities of 20 batches of samples were among 0. 90-1.00. Conclusion The established meth- od of chromatographic fingerprint of Hypericum japonlcum by HPLC is accurate, simple and convenient with good reproducibility, which preliminarily proves the correctness of the Expert System.
