T-AK细胞的形态学和免疫表型研究

摘　　要：ＴＡＫ（Ｔａｃｔｉｖａｔｅｄｋｉｌｅｒ，ＴＡＫ）细胞是由抗ＣＤ３单克隆抗体和ｒＩＬ２共同激活诱生的具有高度溶细胞活性的肿瘤杀伤效应细胞。为探讨ＴＡＫ细胞的免疫生物学特征，研究了ＴＡＫ细胞的形态学和细胞表型。结果表明，ＴＡＫ细胞体积较大，核／浆比例小，胞浆内含有大量颗粒，具有活化淋巴母细胞的形态学特征；９０％以上的ＴＡＫ细胞表达Ｔ淋巴细胞表型（ＣＤ３＋，ＣＤ８＋），２０％～５０％细胞为ＮＫ细胞表型（ＣＤ１６＋，ＣＤ５６＋），并表达ＩＬ２受体、ＣＤ３８和ＭＨＣⅡ类抗原等激活淋巴细胞的表面标志。攻击Ｋ５６２细胞的ＴＡＫ细胞主要为富含颗粒、ＣＤ８＋和ＣＤ５６＋细胞。证明ＴＡＫ细胞是以激活Ｔ淋巴细胞和ＮＫ细胞为主的具有细胞毒活性的异质性细胞群体。おactivated killer (TAK) cells were tumorkilling effectors with high cytolytic activity induced by antiCD3 monoclonal antibody and interleukin2 coactivation. In order to take their biological features the cytomorphology and phenotypes of TAK cells were studied. The results showed that TAK cells were morphologically similar to these of lymphoblasts and more than 90% of TAK cells were phenotypes of T lymphocytes (CD3+,CD8+).Some of TAK cells expressed IL2 receptor, CD38 and MHC Ⅱ antigens characteristic makers for the activated T lymphocytes. TAK cells which attacked the leukemia K562 cells were morphologically large volume with granules, and mainly contained CD8+ and CD56+ cells. The study suggested that TAK cells are heterogeneous cell population with cytotoxicity and consisting of mainly activated T lymphocytes and NK cells.
