摘 要:利用混合淋巴细胞培养反应(MixedLymphocyteReaction,MLR)研究了传统中药当归(AngelicaSinensisDiels,ASD)来源的两种免疫调节剂当归多糖(ASDP)、当归内酯(ASDE3)的免疫作用机理。结果表明:两者均能明显促进由同种异型抗原刺激的正常小鼠及S180荷瘤小鼠脾细胞增殖(P<0.01),其促进作用与剂量之间呈依赖关系。在一定浓度范围内(16~500μg/ml)对未经活化的脾细胞无诱导增殖作用。选择促进效应较大的药物剂量,培养96h,对活化的T淋巴细胞亚群的变化标记检测,结果显示:ASDP可明显增加L3T+4细胞比例,ASDE3可明显增加L3T+4和Lyt+2细胞的数量。在浓度为250μg/ml时,ASDE3增强细胞毒T细胞的功能,其杀伤活性增加80%。おSDP and ASDE3 are extracted from traditional Chinese MedicineAngelica Sinensis Diels (ASD). Their immunopharmacological effects have been studied. Mixed Lymphocyte Reaction was used as a main model through all the experiment. In a series of concentration, ASDP and ASDE3 markedly promoted proliferation of normal mouse and S180 bearing tumor mouse splenocytes stimulated by alloantigens in vitro (P<0.01). The effects were positively correlated with concentration of the agents. In certain range of concentration (16~500 μg/ml), ASDP and ASDE3 could not induce proliferation of inactivated mouse splenocytes. T lymphocyte subsets were assessed after treatment with ASDP and ASDE3 in the optimal concentration, meanwhile take fresh and alloantigen activated lymphocyte as control. It is observed that the L3T+4 cells were increased obviously by treated with ASDP, L3T+4 cells and Lyt+2 cells were also increased markedly by treated with ASDE3 in vitro. The data also show that ASDE3 markedly enhanced the cytotoxicity of cytotoxic T lymphocytes, which was increased 80% at the concentration of 250 μg/ml in vitro.
