摘 要:目的:研究链脲佐菌素(streptozotocin , STZ)对大鼠血糖及胰腺组织内氧化应激影响以及褪黑素( melatonin, MLT)的保护作用。方法:大鼠随机分为三组,正常对照组(NC) , STZ组(STZ) ,褪黑素+STZ组(MLT+STZ),各10只大鼠。STZ(70 mg/kg)一次性腹腔内注射以诱发糖尿病,注射前8天开始给予MLT (200ug/kg/day,强饲)直至实验结束(4周)。实验开始及结束时测动物血糖,称体重各一次,实验结束时处死动物,取胰腺组织行生化检测。结果:STZ使大鼠血糖明显升高,同时升高胰腺组织内MDA水平,降低胰腺组织内GSH水平及GSH-Px活性;MLT预处理降低STZ大鼠的血糖及胰腺组织内MDA水平,增加GSH水平及GSH-Px活性。结论:MLT通过减轻胰腺组织内氧化应激水平保护胰岛β细胞功能的完整性。Objective: To investigate the protective effects of melatonin (MLT) against hyperglycemia and the severity of oxidative stress in pancreatic tissues induced by streptozotocin (STZ ) in rats.Methods: Thirty rats were randomly divided into three groups: Normal control (NC) group,STZ group and MLT﹢STZ group.MLT (200ug /kg/day,oral gavage) was given for 8 days prior to induction of diabetes with STZ (70 mg/kg,i.p.) until the end of the study (4 weeks);Blood glucose levels and biomarkers of oxidative stress in pancreatic homogenates were measured.Results: STZ induced a significant increase in blood glucose levels and malonaldehyde (MDA) levels in pancreatic homogenates,a significant decrease in glutathione (GSH) levels and glutathione peroxidase (GSH-Px) activities.Pretreatment of STZ rats with MLT decreased blood glucose levels,MDA levels and increased GSH levels and GSH-Px activities in pancreatic homogenates.Conclusion: MLT exerts its protective effect against hyperglycemia through reducing the severity of oxidative stress in pancreatic tissues.
