Observation on Stereology of Cell Apoptosis in Cerebral Parietal Cortex after Cerebral Ischemia Reperfusion in the Rats

摘  要:用体视学方法分析大鼠脑缺血再灌注后大脑运动皮质细胞的凋亡情况。随机将24只SD大鼠分为缺血再灌注后6、12、24 h和对照组。采用结扎大鼠左侧颈总动脉和夹闭右侧颈总动脉15m in后放开的方法制备大鼠脑缺血再灌注模型。应用Hoechst33258荧光染色及免疫组化Bax蛋白表达来检测细胞凋亡,用体视学参数定量分析运动皮质区细胞在脑缺血再灌注后不同存活时间内的凋亡情况。结果显示,缺血再灌注后,大脑左右两侧运动皮质Hoechst33258荧光染色以及免疫组化Bax蛋白阳性表达显示的凋亡细胞的体密度和数密度,与对照组比较,缺血再灌注后6、12 h和24 h组均极显著性增大(P<0.01)。以上结果表明缺血再灌注后24 h内运动皮质细胞凋亡逐渐增加,凋亡是脑缺血再灌注后损伤的重要形式之一。To analyze the cell apoptosis changes in parietal cortex after cerebral ischemia-reperfusion in the rats,Twenty-four SD rats were randomly divided into four groups:control group,cerebral ischemia reperfusion of 6 hour,12 hour and 24 hour groups.Cerebral ischemia reperfusion model was performed by ligation of the left common carotid artery and clamping of the right common carotid artery for 15min.The changes of cell apoptosis were observed by Hoechst33258 fluorescein stain and Bax immunohistochemical method,and the quantitative analysis of neuron apoptosis was evaluated by stereology.The result showed that it was distinct increased(P〈0.01)in experimental group comparing with control group that the volume density and numerical density of apoptotic cell showed by Bax positive cells and fluorescence in parietal cortex.This result indicated that the number of apoptotic cells increased with the extention of reperfusion within 24 hours.The cell apoptosis may play an important role in cerebral ischemia reperfusion injury.
