穿透支原体LAMPs诱导小鼠巨噬细胞产生一氧化氮并抑制细胞增殖

摘  要:目的检测穿透支原体脂质相关膜蛋白能否诱导小鼠巨噬细胞产生一氧化氮(NO)及其对细胞增殖的影响,以了解穿透支原体潜在的致病性。方法用SP-4培养基培养穿透支原体并提取其脂质相关膜蛋白,用提取的脂质相关膜蛋白刺激小鼠巨噬细胞,以ELISA法检测NO的产生,并用MTT法检测其对小鼠巨噬细胞增殖的影响。结果穿透支原体脂质相关膜蛋白能以时间和剂量依赖方式刺激小鼠巨噬细胞产生NO,并且对小鼠巨噬细胞的增殖有一定的抑制作用。结论穿透支原体的脂质相关膜蛋白能诱导巨噬细胞产生一氧化氮,且能抑制巨噬细胞的增殖,因而可能与其致病性相关。Objective To investigate the potential pathogenicity of Mycoplasma penetrans (M. penetrans), and to study nitric oxide (NO) production and cell prolification in lipid-associated membrane proteins (LAMPs) -stimulated mouse macrophages. Methods M. penetrans was cultured and its LAMPs were extracted. Mouse macrophages were stimulated with M. penetrans LAMPs to analyze the production of nitric oxide (NO). Cell prolification was detected in M. penetrans LAMPs - stimulated mouse macrophages by MTT assay. Results M. penetrans LAMPs stimulated mouse macrophages to produce nitric oxide in dose and time - dependent manner. And M. penetrans LAMPs inhibited cell prolification in mouse macrophages. Conclusions M. penetrans can stimulate the production of NO and inhibit mouse macrophages prolification, thus it may toe an important etiological factor.
