摘 要:目的研究柴油机尾气颗粒物(DEPs)对小鼠精子质量的影响。方法21~22g健康成年ICR雄鼠随机分为对照组(PBS)、高剂量染毒组、中剂量染毒组、低剂量染毒组,每组6只。每组雄鼠乙醚麻醉后,通过咽后壁吸入的方式分别吸入PBS、360.0、90.0和24.0μg DEPs,每周2次,连续染毒4周,于第6周处死,检测并比较小鼠体重、睾丸及附睾重量,脏器(睾丸、附睾)系数、精子密度、总活力,体外受精率等指标。结果组间睾丸重量、睾丸系数比较差异无统计学意义(P>0.05)。剂量为360.0、90.0μg DEPs的小鼠染毒组与PBS组、24.0μg DEPs小鼠染毒组比较,附睾重量、附睾系数有显著增加(P<0.05);而精子密度、总活力、受精率显著下降(P<0.05)。总活力和受精率之间呈正相关(P=0.000)。结论中、高剂量染毒(360.0、90.0μg DEPs/小鼠)可以降低健康成年雄性ICR小鼠精子质量,低剂量(24. 0μg DEPs/小鼠)则无显著影响。Objective To study the effects of diesel exhaust particles (DEPs) on the quality of mouse sperm. Methods Male mice with twenty one to twenty two grams were randomly divided into high-dose group, middle-dose group, low-dose group and vehicle (PBS) groups (n=6). After anesthesia with ether, ICR mice were treated with PBS 360.0, 90.0, 24.0 ug/mouse DEPs suspension respectively through posterior pharynx, and they were exposed twice a week for four weeks, then were executed at 6th week. Body weights, testis and epididymis weights, ratios of testis weight to body weight, ration of epididymis weight and body weight, sperm density and the total activity, and in vitro fertilization (IVF) rate were detected and analyzed. Results There is no statistical difference among the groups in testis weights or ratios of testis weight to body weight (P〉0.05). The epididymis weights or epididymis weight/body weight in high-dose group and middle-dose group was significantly increased compared with that of low-dose group and PBS group (P〈0.05), wherease sperm density, the total activity, or IVF rate were found to be significantly decreased (P〈0.05). There was a positive correlation between the total sperm activity and the IVF rate. Conclusion High dose DEPs or middle dose DEPs (360.0 and 90.0ug/mouse) might change the quality of mose sperm distinctly, but no changes were found under treatment with low dose DEPs.
