丹参酮ⅡA对人卵巢癌A2780细胞增殖抑制及诱导凋亡的作用

摘　　要：目的研究丹参酮ⅡA(TanⅡA)对人卵巢癌A2780细胞增殖抑制及诱导凋亡的作用。方法以四甲基偶氮唑蓝(M TT)法测定TanⅡA对人卵巢癌A2780细胞的增殖抑制;流式细胞仪检测细胞周期。结果TanⅡA对人卵巢癌A2780细胞的生长具有抑制作用,其作用强度随药物作用时间及浓度的增加而增强。实验组较对照组G0/G1期细胞数量增多,而S期细胞数量减少。4.0mg/L TanⅡA作用0、24、48、72h后的凋亡率分别为4.52%、19.7%、25.3%、40.4%,随时间的延长而凋亡细胞增多。结论TanⅡA对人卵巢癌A2780细胞有明显的增殖抑制和诱导凋亡的作用,且其作用呈时间-剂量依赖性。Objective： To observe the effects of inhibition and inducing apoptosis of tanshinone ⅡA on human ovarian cancer A2780 cell lines. Methods： MTr assay was used to analysis the inhibitive action of tanshinone ⅡA to the A2780 cell line. The cell cycle was determined by flow cytometry. Results： Tanshinone ⅡA could inhibit human ovarian cancer A2780 proliferation, and the intensity increases with time and the concentration of drug. The experimental group had much more G0/G1 phase cells and less S phase cells than the control group. The A2780 cell lines treated by 4.0 mg/L tanshinone HA for 0, 24, 48 and 72 h,and the apoptosis rates were 4. 52%, 19.7%, 25.3%, 40.4%. It showed a time - dependence. Conclusion： Tanshinone ⅡA can significantly inhibit the proliferation of human ovarian cancer A2780 cells and induced the cells apoptosis, and it show a time - dose - dependence.
