摘  要:目的建立测定大鼠灌服清肺口服液后血中白藜芦醇和大黄素浓度的高效液相色谱(HPLC)方法。方法采用H PLC法测定清肺口服液含药血清中白藜芦醇和大黄素中的血药浓度,并对其在大鼠体内的药物动力学特点进行研究。结果白藜芦醇血清质量浓度线性范围为0.075~2.250mg/L,绝对回收率为90.9%~112.2%,相对回收率为87.8%~116.3%;大黄素血清质量浓度线性范围为0.030~1.500μg/ml,大黄素绝对回收率为86.4%~108.2%,相对回收率为85.3%~115.9%;清肺口服液血浆样品中未检出白藜芦醇及大黄素。结论本文建立的测定清肺口服液含药血清白藜芦醇和大黄素血药质量浓度含量的方法简单、灵敏、准确、精密,可有效测定白藜芦醇和大黄素的质量浓度。Objective: To establish a method using HPLC for determination of the concentration of resveratrol and emodin in the blood of rats garaged with Qingfei Oral Liquid. Methods: The plasma concentration of resveratrol and emodin in Qingfei Oral Liquid contained serum was determined by HPLC, then the pharmacokinetic features of resveratrol and emodin in rats were studied. Results: The calibration curve of resveratrol was linear in the range of 0. 075 -2. 250mg/L. The absolute recovery rate of resveratrol was 90.9% - 112.2%, and the relative recovery rate of resveratrol was 87.8% - 116.3%. The calibration curve of emodin was linear in the range of 0. 030 - 1. 500ug/ml. The absolute recovery rate of emodin was 86.4% - 108.2%, and the relative recovery rate of emodin was 85.3% - 115.9%. Resveratrol and emodin were not detected in the plasma sample of Qingfei Oral Liquid. Conclusion: The method that we established for determination of the concentration of resveratrol and emodin in Qingfei Oral Liquid contained serum is simple, sensitive, accurate and precise, and it can be used for determination of the concentration of resveratrol and emodin.
