摘 要:为构建可同时检测单一样品中多种病原体的复合基因芯片,检索相关细菌的16S~23SrRNA间区基因序列,应用生物信息学软件针对不同细菌的共同序列和特异序列,设计通用引物和特异寡核苷酸探针并制成基因芯片。利用芯片对临床泌尿生殖道分泌物样本进行病原菌检测。80例样本中96%芯片检测结果与门诊回复结果符合,反馈的测序结果经BLAST软件对比分析,与其芯片检测结果一致。该泌尿生殖道病原体检测基因芯片设计合理,检测结果具有较好的重复性和可靠性。In order to construct a multi-purposed gene chip for detecting different pathogens in a single sample at the same time, 16S-23S rRNA gene sequences of related bacteria were searched, and universal primer for shared sequences and specific oligonucleotide probes for distinguished sequences were used and designed with bioinformatics software and prepared the gene chip. Using this prepared gene chip to detect pathogens from clinical genitourinary tract secreta was carried out. The results showed that 96% of tested results of 80 cases coincided with out-patient reports; and the feedback sequencing results were identical with comparison and analysis through software BLAST. The design of the gene chip for genitourinary tract pathogens detection was reasonable; the detection results were of fairly good repeatability and reliability.
