摘  要:链霉素经氧-羧甲基羟胺肟化处理后引入羧基,采用EDC一步法分别合成免疫原BSA-EDC-SM和包被抗原OVA-EDC-SM;用戊二醛法和1,4-丁二醇缩水甘油醚分别合成BSA-GA-SM和BSA-di-SM;采用SDS-PAGE凝胶电泳和紫外扫描测得BSA-EDC-SM、BSA-GA-SM和BSA-di-SM的BSA与SM的分子结合比分别为1∶23、1∶15和1∶10。用3种免疫原免疫Balb/C小鼠,获得了高价、敏感和特异的抗血清。BSA-EDC-SM免疫组中1#小鼠的多克隆抗体对SM的IC50为45.8μg/mL,对双氢链霉素(DHSM)的交叉反应为90.6%,与其他氨基糖苷类药物或抗生素药物的交叉反应均小于0.001%。BSA-di-SM和BSA-GA-SM免疫组的多克隆抗体对DHSM、其他氨基糖苷类药物或抗生素类药物的交叉反应均小于0.001%。Immunogen BSA-EDC-SM and coating antigen OVA-EDC-SM were synthesised by using EDC one-step after inleted carboxy group by O-carboxymethyl-hydroxylamine,BSA-GA-SM and BSA-di-SM were synthesized by using glutaraldehyde and 1,4-butanediol diglycidyl ether.Corresponding identification was made by SDS-PAGE and UltraViolet scan.Bind score molecule of BSA and SM of BSA-EDC-SM,BSA-GA-SM,BSA-di-SM were 1∶23,1∶15,1∶10 respectively.The high-titer,sensitive and specific SM antiserum had been produced by immunized 6W Balb/C mice.IC50 of pAb of BSA-EDC-SM immunity class miceⅠ-1# anti-SM was 45.8 μg/mL,cross reaction rate to DHSM was 90.6%,and no cross reaction to other antibiotics.BSA-di-SM、BSA-GA-SM immunity class mice were no cross reaction to DHSM and other antibiotics.
