摘  要:目的探讨快速起搏心房对犬心脏乙酰胆碱M2受体表达和cAMP含量的影响。方法13只犬随机分为两组。在实验组犬左心房缝合电极连接刺激仪持续快速起搏,诱发出持续性房颤,制备犬慢性房颤模型,Western blotting检测实验组和假手术组乙酰胆碱M2受体表达,放射免疫法测定心房cAMP含量。结果实验犬在(25.2±5.2)d内均能诱发出持续超过24h的房颤,实验组M2受体蛋白的表达明显高于假手术组(0.52±0.05vs0.28±0.04,P<0.05),cAMP含量明显下降[(25.7±3.1)pmol/(mg·min)vs(64.8±4.7)pmol/(mg·min),P<0.01]。结论快速起搏心房能明显上调M2受体蛋白的表达,引起cAMP含量下降,表明房颤本身能引起心脏迷走神经的激活,从而有利于房颤的维持。Objective To explore the effects of rapid pacing on exprssion of M2 muscarinic receptor and cAMP in canine atrium. Methods Thirteen dogs were randomly divided into sham group(n=6) and pacing group(n=7).Burst pacing was not performed in sham group.In pacing group,chronic atrial fibrillation models were establish by inducing persistent atrial fibrillation with rapid left atrial pacing(20 Hz).M2 muscarinic receptor was detected by Western blotting and cAMP was determined by radioimmunology. Results The persistent atrial fibrillation more than 24 h was induced by rapid left atrial pacing(20 Hz)in(25.2±5.2)d in pacing group.Compared with sham group,expression of M2 receptor increased significantly in pacing group(0.28±0.04 vs 0.52±0.05,P〈0.05),while levels of cAMP decreased significantly[(64.8±4.7)pmol/(mg·min)vs(25.7±3.1)pmol/(mg·min),P〈0.01]. Conclusion The results suggest that atrial fibrillation induced by rapid pacing could activate cardiac vagus nerve,which could facilitate the maintenance of atrial fibrillation.
