摘 要:目的了解南京地区丙型肝炎病毒(HCV)基因型分布的特点.方法采用针对HCV三种主要基因型1,2,3和1b亚型设计的型-特异性引物,通过RT—PCR和套式PCR扩增HCV基因组高度保守的5’端非编码区,检测55例HCVIgG阳性血清的HCVRNA,并对其中40例进行基因分型.结果PCR检测的抗HCVIgG阳性血清标本55例中,50例为HCVRNA阳性,阳性率为90.91%;对40例HCVRNA阳性的血清进行分型,发现23例为1b型,15例为2,1b混合型,HCV—1b亚型舍有率为95%,其余两例分别为2型和1,2混合型.结论该地区HCV流行优势株为1b亚型.Objective To investigate the distribution of hepatitis C virus(HCV) genotype in Nanjing area. Methods Using type-specific primers for the three major genotypes 1,2,3 and subtype lb of HCV to amplify the 5'NCR that was the most conserved. HCV RNA were tested in serum samples from 155 patients with anti-HCV IgG positive by RT-PCR. Then HCV genotype of 40 patients were analyzed. Results This study found that 50 (90.91%) cases of HCV RNA from 55 patients with anti-HCV IgG positive serum using PCR. Among 40 samples of HCV RNA positive,23 were HCV-1b, 15 were mixtures of HCV-lb and HCV-2,1 was HCV-2,and 1 was a mixture of HCV-1 and HCV-2. Conclusion Determination of HCV genotype by genotype-speeific primer demonstrated that the subtype lb of the HCV is the dominant epidemic strains in Nanjng area.
