摘 要:目的建立一套简单易行的试剂红细胞质量控制检测方案,提高试剂红细胞的质量保证,并为制备试剂红细胞的质量标准提供依据。方法分别对相同方法配制的保存1mo和2mo后的试剂红细胞与新鲜制备的试剂红细胞进行特异性、亲和力和抗原性测定,比较不同保存期试剂红细胞的渗透脆性、变形性、红细胞沉降率、反复冻融抗破坏性。结果保存1mo,2mo的试剂红细胞的特异性、亲和力与新鲜配制的试剂红细胞无明显的差异,但保存2mo后的试剂红细胞的抗原性低于新鲜制备和保存1mo的试剂红细胞,试剂红细胞渗透脆性、变形性和反复冻融抗破坏能力均明显低于新鲜配制的试荆红细胞;保存2mo和1mo后的试剂红细胞自然沉降率明显高于新鲜配制的试剂红细胞。结论试剂红细胞保存至2mo时其特异性、亲和力与保存1mo和新鲜配制的试剂红细胞无差异,可以常规应用,但随着保存时间的延长,其抗原性、渗透脆性、变形性及抗破坏能力都明显下降,在常规应用中应注意。Objective To establish a simple program for quality control of reagent red blood cell in order to elevate quality assurance. Methods To compare the specificity, affinity and antigenicity of reagent red blood cell which were prepared with same method after conserved 1 month, 2 month with freshly prepared ,and compare the osmotic fragility, deformability,blood sedimentation rate,destructiveness of repeat freeze thawing with freshly prepared. Results The specificity,affinity had different change in different keeping time reagent red blood cell,but the red cell antigens of reagent red blood cell which were conserved 2 month were lower than freshly prepared and conserved 1 month. Osmotic fragility, deformability and destructiveness of repeat freeze thawing on conserved 2 month reagent red blood cell were lower than freshly prepared and conserved 1 month. Blood sedimentation rate in conserved 2 month and 1 month were all high than freshly prepared. Conclusion The specificity and affinity of reagent red blood cell after conserved 2 month hadn't difference with freshly prepared,but the antigenicity,osmotic fragility,deformability and destructiveness of repeat freeze thawing were all decreased with the conservation time prolong. It should be carefully to use in routine work.
