摘　　要：目的制备2型肺炎链球菌多糖（Streptococcus pneumoniae type2 polysaccharide，SPNPS2）-破伤风类毒素（tetanus toxoid，TF）结合疫苗（SPNPS2-TY），并研究其免疫原性。方法采用溴化氰活化SPNPS2，已二酰肼作连接剂，碳二亚胺为耦联剂，将活化的SPNPS2与TT耦联制成SPNPS2-TT.以SPNPS2-TT或单纯SPNPS2免疫Balb／c小鼠，用ELISA检测小鼠血清抗SPNPS2 IgG水平.结果SPNPS2-TT的各项指标均达到质控标准。SPNPS2-TT免疫小鼠产生的特异性抗SPNPS2 IgG水平显著高于单纯SPNPS2免疫小鼠，且SPNPS2-TT诱生了免疫记忆应答。结论采用此项技术制备SPNPS2-TT疫苗是可行的。Objective To prepare Streptococcus pneumoniae type 2 polysaccharide （SPNPS2）-tetanus toxoid（TT） conjugate vaccine （SPNPS2-TT） and study its immunogenicity. Methods The SPNPS2 was activated by cyanogen bromide. With adipic acid dihydrazide as linking agent,the SPNPS2-TT was prepared by carbodiimide-mediated coupling of SPNPS2 with TT. The Balb/c mice were immunized with SPNPS2-TT or pure SPNPS2, and the auti-SPNPS2 antibodies were detected by ELISA. Results All the qua-lity indexes of SPNPS2-TT met the standards of quality control. The levels of specific IgG antibody elicited in the mice immunized with SPNPS2-TT were significantly higher than those in the mice immunized with pure SPNPS2, and only SPNPS2-TT induced immune memory. Conclusion The SPNPS2-TT is successfully prepared with this technology.
