摘 要:采用犬双后肢低、高速投射物致伤模型,探讨了伤后早期边缘系下丘脑、海马区及颞顶区灰、白质脑组织内皮素-1(ET1)表达与释放及其超微结构改变。结果发现,伤后8h高速组下丘脑、海马区ET1含量明显增高(P<0.01);低速组下丘脑,高速组下丘脑、海马区ET1-mRNA表达增强(P<0.05);超微结构改变仍以高速组下丘脑、海马区为主,多集中于神经元、神经纤维及毛细血管内皮细胞的变化。提示伤后脑组织ET1释放表达增强,以及边缘系下丘脑、海马区选择性易损,可能为创伤后应激障碍早期重要病理生理基础之一。With the stress model of low and high velocity projectiles injury to hind thighs in dogs, the expression and release of endothelin1(ET1) were observed in limbic system and temporoparietal grey and white matters, together with the early pathomorphism changes of their ultrastructure. The results showed that ET1 levels markedly increased in hypothalamus and hippocampus in high speed group after 8 hours of posttrauma(P<001); the expressions of ET1mRNA in hypothalamus in low speed group, hypothalamus and hippocampus in high speed group were significantly enhanced(P<005). The early pathomorphism changes displayed some alterations in neurons, nerve fibers and capillary endothelium cells of limbic system in high speed group. It was suggested that the increased expressions of ET1 in brain tissues and the selectively vulnerable damage of hypothalamus and hippocampus might play a consistent pathophysiological role in the early period of posttraumatic stress disorder.
