巨噬细胞条件培养基影响新生鼠雪旺氏细胞表达神经生长因子的实验研究

摘  要:为了研究巨噬细胞对雪旺氏细胞表达神经生长因子的影响,制备了脂多糖激活的和未激活的成年大鼠腹腔巨噬细胞的条件培养基,取3种不同浓度,分别施加于纯化培养的新生鼠雪旺氏细胞。作用24、48、72h,用神经生长因子单抗及间接免疫荧光法显示细胞,用流式细胞仪进行测试和统计分析。结果发现,脂多精激活的巨噬细胞条件培养基或未激活者,取其不同浓度、作用不同时间后,均能不同程度地提高新生鼠雪旺氏细胞的神经生长因子阳性表达率及细胞内的神经生长因子含量。提示巨噬细胞条件培养基中含有促进雪旺氏细胞表达神经生长因子的物质,巨噬细胞可能通过分泌细胞因子调节雪旺氏细胞对神经生长因子的表达.In order to study the innuence of macrophages on HGF (nerve growth factor) expression in newborn rat Schwann cells, LPS-activated and not-activted adult rat peritoneal macrophages conditioned media were cultured with purified newborn rat Schwann cells for 24, 48 and 72 h in three different concentration, then marked Schwann cells with anti-NGF antibody by indirect immunonuqresent methods, and measured the NGF-positive-expression-percentage and cellular NGF contents in Schwann cells by now cytometer, The results indicated that both L-MocM and N-MocM enhanced the Schwann cells expressing NGF rate and cellular NGF contents in different concentration or testing time. It is confirmed that MocM contained some factors which can increase NGF-expression in Schwann cells in vitro, and implied that macrophages performed the effect by secreting some cytokines.
