摘  要:目的应用表面加强激光解吸电离飞行时间质谱(SELDI-TOF-MS)技术分析急性重度颅脑损伤病人的差异性蛋白质谱,评价此技术对颅脑损伤蛋白质组学的检测价值及临床应用前景。方法应用SELDI-TOF-MS技术比较创伤组和对照组脑组织之间、血清之间蛋白质的差异表达,筛选出血清、脑组织之间相同的差异蛋白质。结果共有116个蛋白质波峰质/核比(M/Z)在2000~20000之间被标记,其中101个M/Z集中在2000~10000;创伤病人脑组织、血清之间筛选出8个相同的差异性M/Z:3178、3412、4201、4692、8605、8676、9387、9607。结论SELDI-TOF-MS是一种新的蛋白质组学技术,能够检测样本中大量的低分子量蛋白质,具有快速、简便易行且高通量的特点,可直接筛选颅脑损伤的标记物,有较好的临床应用前景。Objectives To analyze the mass spectrum of differential proteins by surface enhanced laser desorption/ianization time of flight mass spectrum (SELDI-TOF-MS) proteinChip technology, and to estimate its clinical significance in the patients with traumatic brain injury (TBI). Methods The mass spectra of protein in the cerebral cortex tissues and serum were determined by SELDI-TOF-MS proteinChip technology in both the experimental and control groups. The differential proteins between both the experimental and control groups were screened out. Results There were 116 protein peaks, of which, ratio of mass to charge varied from 2000 to 20000, in the cerebral tissues and serum of the patients with TBI. The ratio of mass to charge in 101 protein peaks varied from, 2000 to 1000@ Eight peaks of the same proteins in the cerebral tissues and serum of the experimental group were different from those of the control group. The ratios of mass to charge of 8 differential proteins were 3178, 3412, 4201, 4692, 8605, 8676, 9387, and 9607 respectively. Conclusions SELDI-TOF-MS is a quickly, easy and convenient, and high flux proteomics technology, which is capable of screening differential and potential biomarkers from acute severe TBI patients, and perhaps will clinically be better applied in future.
