摘  要:目的:探讨黄芪皂苷(AST)抑制HEK293sw细胞-淀粉样肽(Aβ)生成的作用机制。方法:采用HEK293sw细胞,常规培养后加入不同浓度(0,0.1,1和10μmo.lL-1)AST处理,荧光法测定β-分泌酶活力,并用Western blot和RT-PCR技术检测淀粉样前体蛋白(APP)和β-位点APP切割酶1(BACE1)的表达。结果:在HEK293sw细胞模型上,AST终浓度1和10μmol.L-1显著降低β-分泌酶活力。1和10μmol.L-1AST对APP的蛋白和mRNA表达无影响,但对BACE1的蛋白水平都有降低作用,同时也显著降低mRNA的水平。结论:AST可能通过抑制β-分泌酶而降低Aβ的产生。Objective: To investigate the mechanism of the inhibiting effect of AST on β-amyloid peptide(Aβ) production in HEK293sw cells.Methods: Routinely cultured HEK293sw cells were treated by different concentrations of AST.At 72h later,cells were examined for β-secretase activity by fluorescence method with a kit supplied by R&D Systems,and were examined for amyloid precursor protein(APP) and β site APP cleaving enzyme 1(BACE1) protein expressions by Western blot and for mRNAs with RT-PCR.Results: At 1 and 10μmol·L^-1,AST significantly decreased β-secretase activity,had no effect on protein or mRNA expression of APP,but decreased BACE1 protein and mRNA level in a concentration-dependent manner.Conclusion: AST can decrease the expression of Aβ protein through the inhibition of β-secretase in HEK293sw cells.
