甲状腺激素对仔鼠被膜下胸腺上皮细胞影响的超微结构观察

摘  要:【目的】研究给予甲状腺激素后,仔鼠被膜下胸腺上皮细胞超微结构的变化。【方法】取300 g左右的健康Wistar大鼠,交配后,雌性大鼠随机分为2组:对照组和实验组。各组均在发现阴栓后15 d灌胃,直到仔鼠生后20 d。实验组每只大鼠给予浓度为50 g/L的甲状腺素混悬液1 ml/d,对照组每天给予等量生理盐水。两组分别在仔鼠生后的1、10、20 d处死,取胸腺,电镜观察被膜下胸腺上皮细胞的超微结构;采用放免法测定仔鼠血清FT3和FT4水平。【结果】与对照相比,实验组10 d龄仔鼠的FT3显著升高,其余未见明显异常。实验组仔鼠胸腺上皮细胞发生了明显变化:细胞排列松散,间隙增宽;细胞水肿;凝固性坏死;胞膜溶解。【结论】在胸腺的发育过程中,过量的甲状腺激素会严重损伤胸腺上皮细胞。[Objective] To investigate the uhrastructaral changes of subcapsular thymic epithelial cells in newborn rats after mother rats administered with thyroid hormones. [Methods] The female Wistar rats (weigh about 300 g) were divided randomly into two groups after vaginal plug were found: normal group and experimental group. Rats were administered by gavage from the 15th gestational days to 20 days after birth, with lml/d thyroid hormones suspension (concentration 50 g/L) in experimental pregnant rats and physiological saline in normal group. Newborn rats were sacrificed in both group on 1, 10, 20 days after birth and subcapsular thymic epithelial cells were observed by electron microscopy. The level of thyroid hormones FT3 and FT4 was measured by radioimmunoassay [ Results] The FT3 level in experimental group on 10th days was higher than that in normal group ( P 〈 0.05) ; there were no dramatically changes in other indices. Subcapsular thymic epithelial cells appeared significant changes on ultrastructure: cells arranged loosely, intercellular space broaden, with cellular swelling, coagulative necrosis and lysis of cell membrane. [Conclusions] During thymus development, Excessive thyroid hormones intaking by mother rats may injure the subcapsular thymic epithelial cells of newbom rats.
