摘  要:【目的】探讨玻璃化保存法对兔肌腱组织形态学的影响。【方法】取24条新西兰纯种大白兔胫前肌肌腱作为实验对象,随机分为3组,每组8条肌腱:未处理(A)组,"两步法"深低温冷冻保存组(B)组,玻璃化保存(C)组。透射电镜观察肌腱组织细胞的形态,台盼蓝染色法检测胫前肌腱细胞存活率。【结果】A组与C组电镜观察结果类似,细胞的结构形态未见明显改变,B组肌腱组织微观结构损伤明显,经统计学处理,胫前肌腱细胞存活率:C组(89.4±3.7)%与A组(93.3±1.7)%无统计学意义(P>0.05),与B组(74.3±4.8)%比较有统计学意义(P<0.05)。【结论】与"两步法"深低温冷冻保存法相比,玻璃化保存法对细胞损伤小,保存效果较好,随着冷冻保护剂的改进,玻璃化保存法保存肌腱具有良好的发展前景。[Objective] To investigate the effect of vitrification on the histomorphology of tendon tissue in rabbit. [Methods] A total of 24 tendens of New Zealand rabbit were randomly divided into 3 groups and treated, respectively: with cryopreservation by vitrification (C group, n = 8), cryopreservation by two-step method (B group, n = 8), or left untreated as control (A group n = 8). Transmission electron microscope was used to observe the hislogy of tendon. Survival rate of tendon cell was detennined by trypan blue method. [ Results] There were no significant difference between the groups of fresh tendon and vitrified tendon. And no apparent change appeared, but wc also could see that the tissuc's micmmcchanism were apparently damagdd in cryoprcscrvation group. [Conclusions] The damage of vitrifieation to tendon is less than that of profound hypothennia preservion, especially to the corpuscular shape of tendon, and there is a better prospect in the method of vitrification with the development of cryopreserved agent.
