摘  要:目的探讨川芎嗪(tetramethylpyrazine,TMP)对去卵巢大鼠骨质的保护作用及其作用机制。方法将48只雌性SD大鼠随机分成6组,分别为A组:假手术(Sham)组;B组:去卵巢(OVX)组;C组:OVX+TMP小剂量组;D组:OVX+TMP中剂量组;E组:OVX+TMP大剂量组;F组:OVX+雌激素干预组。C、D、E三组于术后1周开始分别行TMP50mg/kg、100mg/kg和200mg/kg剂量腹腔注射;A、B两组分别予同体积生理盐水,每日1次腹腔注射;F组予苯甲酸雌二醇(1mg)隔日注射,疗程均为3个月。分别对其血清碱性磷酸酶(ALP)、血小板活化因子(PAF)、抗酒石酸盐酸性磷酸酶(TRACP-5b)及腰椎椎体、股骨骨密度等进行检测,对腰椎椎体骨行HE染色及核转录因子kappaB(NF-κB)免疫组化染色。结果 (1)与B组相比,其他各组大鼠血清PAF、ALP、TRACP-5b等均呈不同程度的降低(P<0.01-0.05),而腰椎及股骨骨密度值指标呈不同程度的升高(P<0.01-0.05);相比之下,C、D、E三组尤其是E组的上述参数指标与B组相比差异呈现出明显的统计学意义(P<0.001);(2)与B组相比,其他各组NF-κB的表达灰度值呈不同程度的降低(P<0.001-0.05),尤以E组的上述参数指标与B组相比差异呈现出明显的统计学意义(P<0.001);(3)通过图片结果可以清晰地看到腰椎骨小梁变细、断裂、间隙增宽、数量减少,而椎体骨小梁趋于正常,骨小梁间隔匀实,接近Sham组。结论初步证实TMP对去卵巢大鼠骨质具有一定的保护作用,且该作用与剂量相关,这种保护作用可能与减少PAF生成及阻断PAF与PAFR结合和降低NF-κB的表达有关且该保护作用与雌激素干预作用相似,为我们防治绝经后骨质疏松症提供了一个新的靶点。Objective To observe the influence of tetramethylpyrazine(TMP)on bone quality of ovariectomized rats,and to study the possible mechanism.Methods The animal model was established with female SD rats.The 48 SD rats were randomly divided into 6 group:group A:sham operation(sham)group;group B:ovariectomized(OVX)group;group C:OVX treated with small dose TMP group;group D:OVX treated with mean dose TMP group;group E:OVX treated with large dose TMP group;group F:OVX treated with ERT group,with 8 animals in each group.From the one week after operation,group A and group B was performed intraperitoneal injection with normal saline 2 ml every day;group C was performed intraperitoneal injection with TMP 50 mg/kg every day;group D was performed intraperitoneal injection with TMP 100 mg/kg every day;group E was performed intraperitoneal injection with TMP 200 mg/kg every day;group F was performed intramuscular with Estradiol Benzoate(E2)1 mg EOD,the course of treatment was 4 weeks,8 weeks,12 weeks,the ALP,TRACP-5b,PAF and bone density were tested,and the histomorphology of bone was disposaled by HE staining and the NF-κB expression by immunohistochemistry staining.Results(1)The PAF,ALP,TRACP-5b of OVX group were higher than sham group(P<0.01-0.05).The bone density of OVX group were lower than sham group(P<0.01-0.05).All parameters referred in therapeutic groups presented significant differences compared with those in OVX group(P<0.001),large dose TMP therapeutic group especially.(2)The gray scale of NF-κB expression was higher in OVX group than in sham group(P<0.01),and the bone trabeculae in OVX group became thinner and gaps became wider compared with sham group;the gray scale of NF-κB expression was lower in therapeutic groups compared with OVX group(P<0.001-0.05).All parameters referred in therapeutic groups presented significant differences compared with those in OVX group,and the bone trabeculae in therapeutic group became thick and gaps became smaller and bone microstructure improed significantly compared with OVX gro
