摘 要:目的探讨酯酶(EST)同工酶酶谱特征在蚋类分类上的价值,并为蚋类系统发育的研究积累生物化学方面的基础资料。方法应用垂直板不连续聚丙烯酰胺凝胶电泳技术,对素木蚋、双齿蚋和五条蚋进行EST同工酶电泳,测量并计算各酶带的迁移率。结果3蚋种的EST同工酶酶谱共显示10条酶带,种间存在明显差异,具有各自的特征性酶谱,种内个体之间、幼虫与蛹之间酶谱主带相同,非主带略有差异。结论EST同工酶酶谱可作为蚋种鉴定的特征之一。Objective To explore the value of isozyme zymograms in the classification of blackflies and accumulate basic biochemical data for the study of phylogcncsis. Methods EST isozymes of three blackfly species were studied by discontinuous PAGE techniques. Results Ten isozyme hands could be identified according to the electrophoresis zymograms. The differences of zymograms were significant between three blackfly species and each species had its own special zymogram. The main band among intraspecific individuals or between the larvae and pupae was alike, however, other enzyme bands were different. Conclusion EST isozyme zymograms could be used as one of the classification characteristics of blackflies.
