摘  要:从蚯蚓体内分离纯化30 kD以下的抗菌肽,并研究其抑菌效果.通过电击诱导蚯蚓产生抗菌肽、超滤浓缩脱盐以及截取3~30 kD的抗菌肽、用阳离子交换分离除去较多杂蛋白、凝胶过滤层析进一步精确分离提纯得到3~30 kD的蚯蚓抗菌肽,有H1和H2两个蛋白浓度较高的峰值.测得其蛋白质量浓度分别为6.82、7.46 mg/μL,其相对分子质量分别在5 856~7 823 kD和14 40~20 00.抑菌活性测定结果表明,H1和H2都有较好的抑菌效果,其中H1对草枯杆菌、金黄色葡萄球菌、巴氏杆菌、大肠杆菌有明显的抑菌效果,H2对巴氏杆菌、大肠杆菌和草枯杆菌也有明显的抑菌效果,H1的抑菌活性比H2高.In order to isolate and purify the 3 ~30 kD antimicrobial peptides from earthworm and examine its antibacterial activity.The antimicrobial peptide of earthworm were produced by lightning stroke,and isolate the 3~30 kD antimicrobial peptides by ultrafiltrating,concentration and desalinization.Using the SP-Sepharose Fast Flow ion exchang chromatography to remove most of the hybridprotein and Sephadex G-50 gel filtration chromatography to precisely abstracte 3~30 kD antimicrobial peptides which have two wave crest of protein named H1,H2.Their proteinic concentration are 6.82 and 7.46 mg/μl and the molecular mass are between 5.856~7.823 kD and 14.4-20.10 kD.The antimicrobial tests reveal that the antibacterial activity of H1 and H2 is great.Antibacterial activity of H1 on Bacillus subtilis,Staphyloccocus aureus Rosenbach,Pasteurella,E.Coli is obvious.While the antibacterial activity of H2 is good on Pasteurella,E.Coli,Bacillus subtilis.In addition,The antibacterial activity of H1 is better than H2.
