摘 要:目的探讨肝乐颗粒对四氯化碳诱导的肝纤维化大鼠肝组织α-平滑肌肌动蛋白(α-SMA)表达的影响。方法除正常组大鼠外,其余各组大鼠采用四氯化碳诱导肝纤维化模型。于造模第7周起,给药组分别灌胃给予相应的受试药物,疗程6周。实验结束后,采用分光光度法测定血清一氧化氮(NO)、一氧化氮合酶(NOS)和诱导型一氧化氮合酶(iNOS)的含量;同时以免疫组化技术测定肝组织中α-SMA蛋白的表达,RT-PCR测定肝组织中α-SMAmRNA的表达。结果肝乐颗粒各剂量组能明显降低肝纤维化大鼠NO、NOS、iNOS的含量,抑制肝纤维化大鼠肝组织中α-SMA蛋白和α-SMAmRNA的表达。结论肝乐颗粒能有效抑制四氯化碳诱导的肝纤维化大鼠肝脏中α-SMA的表达。Objective To observe the change of α-smooth muscle actin (α-SMA) expression in carbon tetrachloride (CCl4)-induced liver fibrosis in rats after treatment with Ganle keli. Methods Liver fibrosis rat model was induced by CCl4 twice a week for 12 weeks. Ganle keli was used daily intragastric administration from the 7th week. Contents of NO, NOS and iNOS were determined by absorption spectrometry. Liver samples were collected after experiments and stained by immunohistochemistry of α-SMA. Moreover α-SMAmRNA expression in liver tissue was detected by RT-PCR technology. Results Ganle keli could significantly reduce NO, NOS and iNOS contents, effectively prevent the expression of α-SMA, and significantly reduce α-SMA mRNA in liver fibrosis rats. Conclusion Ganle keli can effectively inhibit the expression of α-SMA in CCl4-induced fibrotic liver in rats.
