兔眼玻璃体腔注射贝伐单抗的视网膜毒性研究

摘  要:目的研究不同剂量贝伐单抗(商品名:阿瓦斯汀,avastin)玻璃体腔注射后对动物视网膜组织结构和功能有无毒性并探讨安全剂量。方法32只健康成年有色家兔随机分为3个实验组和1个对照组。实验组每只动物右眼玻璃体腔分别注射avastin 4.2 mg/0.2 mL,0.42 mg/0.2 mL,0.042 mg/0.2 mL,对照组每只动物右眼玻璃体腔分别注射等量的平衡盐溶液。注药后观察眼底改变;第7、14天取眼球,在光学显微镜及透射电镜下观察视网膜组织结构变化。结果对照组及0.42 mg组、0.042 mg组眼底检查及光镜、电镜检查未见异常;4.2 mg组眼底相片显示个别动物出现玻璃体混浊,光镜、电镜检查第7、14天可见视网膜结构改变。结论玻璃体腔注射0.42 mg以下(含0.42 mg)avastin,在短期内对动物视网膜组织结构和神经传导功能无显著影响。Objective To study whether it is toxic for animal's retinal tissue and function,when injected differ- ent dosage of avastin to its vitreous chamber and discuss the safe dosage. Methods Thirty-two healthy color adult hares are divided at random into three experimental groups and a control group.In the experimental groups,each animal's right eye vitreous chamber was respectively injected avastin 4.2 mg/0.2 mL,0.42 mg/0.2 mL,0.042 mg/0.2 mL. In the centrll group each animal's right eye vitreous chamber was respectively injected equal amount of balanced salt soludon. After injection, we observe fundus change and observe retinal tissue structure change of 7^(th) day, 14^(th) day eye hall under optical microscope and transmission electron microscope.Results 0.42 mg/0.2 mL group,0. 042 mg/0.2 mL group,optical microscope and electron microscope examination both appearnomal.In 4.2 mg/0.2 mL group,eye ground pictures show that one or two animals appear vitreous opacity.Retinal structure change may be seen at 7^(th) day,14^(th) day under optical microscope and electron microscope.Conclusion Vitreous chamber was injected avastin below 0.42 mg/ 0.2 mL(including 0.42 mg/0.2mL),in the short term,it doesn't affect animal's retinal tissue structure and nerve conductive function.
