新型口服四价铂族抗癌药赛特铂原料药及其杂质的高效液相色谱测定

摘　　要：赛特铂是一新型、可以口服给药的四价铂类金属抗癌药物。本文采用反相高效液相色谱法外标法:ODS色谱柱(150mm×4.6mm i.d.,5μm),以乙腈:水(20～25:75～80 V/V)为流动相,流速0.8mL/min,在210 nm处检测,成功实现了赛特铂及其杂质的分析测定。该方法测定赛特铂及其杂质JM149、JM118的线性相关系数分别为:0.999、0.998、1.000,线性相关;最小检出限分别为0.932μg/L、0.396μg/L、5.20μg/L;各物质间的分离度均>1.5;赛特铂的回收率:99.2%～100.4%;精密度为0.55%。该分析方法已成功地应用于赛特铂的生产和研究中。Satraplatin is a novel oral platinum(Ⅳ)anticancer drug which has been separated and determined from its impurities under the conditions of a column ODS(150mm×4.6mm i.d.,5μm),acetonitrile-water (20～25:75～80,V/V)as mobile phase,flow rate 0.8mL/min,and detection at 210nm.This method de- tect the linear relative coefficient of Satraplatin and its impurities Jm149,Jm118:0.99,0.998,1.000;the linear is relastive.The minimum detection is 0.932μg/L,0.396μg/L,5.20μg/L,separately;the resolution of all substances are more than 1.5,recovery rate of Satraplatin is 99.2%～100.4%,accuracy 0.55%.The a- nalysis method have been successfully used in production and research of Satraplatin.
