摘　　要：目的探讨粒细胞集落刺激因子(G-CSF)动员正常供者外周血造血干细胞的效果、毒副作用及影响因素。方法对59名异基因造血干细胞正常供者采用 G-CSF 皮下注射3～5d,使用 COBESpectra 血细胞分离机采集外周血干细胞,流式细胞术检测采集物中 CD_(34)^+细胞数。结果所有供者第一次采集的单个核细胞(MNC)及 CD_(34)^+细胞量平均值分别为4.4(1.12～13.06)×10~8/kg 供者体重及3.78.(1.14～12.92)×10~6/kg 供者体重。患者不良反应轻微。男性供者、年龄小于45岁者及采集前白细胞计数高者采集所得 CD_(34)^+细胞数较高。结论 G-CSF 作为正常供者动员剂安全有效。患者性别、年龄及采集前白细胞计数可作为预测因素。Objective To study the efficacy,safety and predictable factors of G-CSF-based mobiliza- tion in normal healthy donors for allogeneic peripheral blood stem cell(PBsc)transplantation.Methods G- CSF was used to mobilize PBSCs in 59 healthy donors and PBSCs were collected by COBE Spectra cell sepa- rator.CD_(34)^+cells were calculated by flow cytometry.Results The numbers of mononucleated cells(MNCs) and CD3_(34)^+cells of the first collection were 4.4(1.12～13.06)×10~8 and 3.78(1.14～12.92)×10~6 per kilogram donor weight,respectively.The side effects were mild.Significantly more total number of CD3_(34)^+cells were mobilized in men,<45 years old or those with higher leukocyte counts before apheresis.Conclusion G-CSF mobiliza- tion of PBSCs in normal healthy donors is effective and safe.Sex,age or leukocyte counts before apheresis can predict CD_(34)^+cell collections.
