摘  要:目的研究培养在壳聚糖-明胶膜片材料上黑色素细胞的生物学特性。方法取幼儿包皮,酶消化成单细胞悬液,接种后在黑色素细胞条件培养液中静置培养。黑色素细胞纯化、扩增后接种于壳聚糖-明胶膜片上。定期观察细胞形态变化及生长增殖情况,应用MTT测定、黑色素瘤抗体(Melan-A)免疫荧光染色、DOPA反应以判断黑色素细胞在壳聚糖-明胶膜片上的增殖及分化情况。结果分离、纯化的黑色素细胞接种于壳聚糖-明胶膜片上生长良好,Melan-A免疫荧光染色及DOPA反应为阳性。结论壳聚糖-明胶膜片是支持人黑色素细胞生长的良好材料,这一体系为色素脱失症的治疗提供了新的途径。Objectives To study biological performance of melanocytes cultured on chitosan-gelatin membrane (CGM). Methods Epidermal keratinocytes contain melanocytes were harvested from human foreskin under informed consent. Melanocytes were purified and enriched in the condition culture medium (Medium 154, M154) and then seeded onto CGM. The cell was cultured on CGM and studied with histology, MTT, Immunofluorescence (melan -A), and L dihydroxyphenylalanine reactivity. Results Melanocytes could be purified and enriched in culture medium 154. Melanocytes cultured on CGM were grew well, Melan-A staining and L-dihydroxyphenylalanine reactivity is positive. Conclusions We have found chitosan-gelatin membrane is a good material support growth of melanocytes. This system may provided an alter approach for treatment of pigment loss.
