摘  要:目的:对自体及异体皮肤成纤维细胞和肌腱细胞的生长因子在分子水平的表达差异进行研究。方法:体外培养自体及异体人肌腱细胞和人皮肤成纤维细胞,用基因芯片的方法对两种细胞的生长因子进行检测及比较,并用RT-PCR的方法进一步验证。结果:自体皮肤、肌腱细胞生长因子的分子水平表达无差异,而异体皮肤、肌腱细胞生长因子分子水平的表达略有差异。结论:自体皮肤成纤维细胞与肌腱细胞的生长因子表达相似,证实两种细胞具有同源性,自体皮肤成纤维细胞可以成为构建组织工程化肌腱的种子细胞。而异体皮肤成纤维细胞和肌腱细胞的生长因子表达略有差异,可能为个体差异所致。Objective: To study the differential gene expression of growth factors between humantenocytes and dermal fibroblasts derived from same or different individuals. Methods: Humantenocytes and dermal fibroblasts were cultured in vitro and gene expression comparison was madebetween the cells using gene chip and RT-PCR. Results: All growth factors investigated in humantenocytes and dermal fibroblasts derived from same individual were similarly expressed. However,differential expression of TGF-RII and IGFBP3 was observed in human tenocytes and dermal fibro-blasts from two patients. Conclusion: The growth factors are similar between human tenocytes anddermal fibroblasts of the same individed. Thus, autologous dermal fibroblasts may potentially beused in tendon engineering as seed cells. The differential gene expression observed between hu-man tenocytes and dermal fibroblasts derived from two patients may be caused by individual varience.
