摘 要:目的观察加替沙星与地高辛合用后其对地高辛血药浓度和药代动力学参数的影响,为临床安全、有效、合理使用强心苷类药物提供参考依据。方法用荧光偏振免疫法测定不同给药方法中家兔体内地高辛血药浓度,并对结果进行统计分析。结果合用加替沙星后,地高辛的血药浓度明显增高,地高辛在家兔体内的分布半衰期(T1/2)延长,表观分布容积(Vd)增大(P<0.05),而血浆清除率(CL)和曲线下面积(AUC0~∞)无显著变化(P>0.05)。结论加替沙星可显著增加地高辛的血药浓度,增加地高辛在体内的蓄积,两者合用时应及时监测地高辛血药浓度,制定个体化给药方案,确保临床疗效,减少毒性反应的发生。Objective To investigate the effects of Gatifloxacin on plasma levels and pharmacokinetics of digoxin in rabbits and to provide evidences for safety, efficacy and proper use of cardiac glycosides. Methods The plasma level of digoxin in rabbits was determined by fluorescence polarization immunization.The results were subjected to statistical analysis. Results The plasma digoxin level, half-life (T1/2) and apparent volume of distribution(Vd)were significantly elevated (P〈0.05), but the plasma clearance (CL) and the area under side curve (AUC0-∞)remained unchanged (P〉0.05) when single dose of gatifloxacin was given with digoxin. Conclusion Gatifloxacin was shown to increase the plasma level and accumulation of digoxin in rabbit. To ensure clinical efficacy and reduce incidence of toxic reactions, it is of great importance to monitor the plasma digoxin level and individualize dosage regimen when the two drugs are used together.
