摘 要:目的 观察丹参酸乙对大鼠肝星状细胞α-平滑肌肌动蛋白的影响。方法 原位灌注、消化法分离正常大鼠肝星状细胞,异硫氰酸胍一步法提取细胞总RNA,RT-PCR法检测α-平滑肌肌动蛋白基因表达,免疫印迹法分析α-平滑肌肌动蛋白量。结果 10μmol/L SAB可抑制肝星状细胞α-平滑肌肌动蛋白的基因表达,但不能抑制其蛋白表达。细胞经TGFβ1刺激后,不仅1μmol/L SAB甚至10μmol/L SAB均不能抑制a-平滑肌肌动蛋白的基因表达。结论 丹参酸乙在肝星状细胞活化的启动阶段可延缓其活化,对于已活化的细胞丹参酸乙则无逆转作用。Objective To observe the effects of salianic acid B on smooth muscle a-actin in rat hepatic stellate cells.Methods Hepatic stellate cells were obtained from normal rat by perfusion and digestion in situ.Total RNA was extracted by acid guanidium single-step method.Gene expression of smooth muscle a-actin was assayed by RT-PCR.Immunoblorting was used to observe the expression of smooth muscle a-actin protein.Results 10μmol/LSAB could inhibit gene expression of smooth muscle a-actin in rat hepatic stellate cells,but could not inhibit its protein.Not only 1μmol/LSAB but also 10μmol/LSAB couldn' t suppress gene expression of smooth muscle a-actin in transforming growth factor β1-stimulated rat hepatic stellate cells. Conclusion Salianic acid B could retard activation of hepatic stellate cells in the initiating stage of activation, but could not affect the activated hepatic stellate cells.
