益舒软肝丸对肝硬化大鼠TGF-β1及TIMP-1表达的影响

摘  要:目的研究益舒软肝丸抗肝硬化的作用机理。方法以CC l4复合因素法建立肝硬化模型,首先制备出肝硬化的实验动物模型,造模结束后,连续灌胃给药3个月。检查ALT、AST,A/G、CHE;HA、LN、PⅢNP、Hyp;TGF-β1和TIMP-1。结果与模型对照组大鼠比,用药组组织切片显示汇管区有少量纤维组织增生,但未见假小叶结构;肝功能指标ALT、AST减少,A/G、CHE增加,肝功能有显著好转;HA、LN、PⅢNP、Hyp水平显著降低,肝纤维化程度降低;TGF-β1和TIMP-1阳性表达减少。结论益舒软肝丸有明显的抗肝硬化的作用,其作用机制可能与下调大鼠组织TGF-β1和TIMP-1的表达,抑制细胞外基质的合成和胶原增生、沉积有关。Objective To investigate the effect and mechanism of Yishuruanganwan(YS) on liver fibrosis induced by CCl4. Methods 80 Wistar rats were randomly divided into normal group model group and three YS groups (0.4, 0.8 and 1.6 g/kg). The model of liver fibrosis was induced by CC14 and high-fat diet in rats. After treatment with drug for 3 months, the histopathological change of liver was observed, the levels of ALT, AST, HA, LN, PⅢNP in serum, and the levels of A/G, CHE and Hyp in liver tissue were determined, and the expressions of transforming growth factor beta (TGF-β1) and the tissue inhibitor of metalloproteinasc-1 (TIMP-1) was detected. Results Compared with model group, YS could significantly alleviate the histopathological injury of liver induced by CCl4, decrease the levels of ALT, AST, HA, LN and PIII NP in serum, increase the levels of A/G, CHE, and reduce the content of Hyp in liver tissue. In addition, the expression of TGF-β1 and TIMP-1 in YS groups was markedly decreased as compared to model group ( P 〈 0.05). Conclusion YS has obvious protective effect on cirrhosis of liver and the mechanisms may be related with the decrease of TGF-β1 and TIMP-1 expression and the inhibition of extracellular matrix synthesis.
