摘  要:为研究不同检测体系下骨髓源间充质干细胞(MSC)迁移特征的差异,本研究利用Boydenchamber观察hSDF-1对MSC迁移的影响是否存在浓度依赖性;观察上述体系经50nmol渥曼青霉素(wortmannin),10μmol/LLY294002,50μmol/LPD98059,10μmol/LU73122,126μmol/LAMD3100以及50nmol/L维拉帕米等处理MSC后对MSC迁移的影响。结果表明:MSC迁移能力随着hSDF-1α浓度的递增而逐渐增强,并且发现Wortmannin、LY294002、PD98059、U70312、AMD3100、Verapamil对MSC迁移均有影响,其中U73122、AMD3100对MSC迁移阻断的效应最显著。结论:SDF-1/CXCR4所介导的MSC迁移可能与丝裂原活化蛋白激酶(MAPK)、磷脂酰肌醇特异性磷脂酶C(PI-PLC)和蛋白激酶C(PKC)等信号途径有关。The aim of this study was to explore the difference of MSC migration mediated by SDF-1/CXCR4 axis through Boyden chamber in vitro migration assay. The SDF-1 density-dependence of MSC migration was observed. Sequently, the effects of different bloking agents on hSDF-MSC migration were observed after MSC were treated with 50 nmol/L wortmannin, 10μmol/L LY294002,50μmol/L PE08059,10μmol/L U73122, 126 μmol/L AMD3100 and 50 nmol/L verapamil respectively. The results showed the efficiency of MSC migration increased gradually with the increasing of hSDF-1 density. And after MSCs treatment with 50 nmol/L wortmannin, 10 μmol/L LY294002,50 μmol/L PD98059,10μmol/L U73122 and 126 μmol/L AMD3100 respectively, the ability of MSC migration decreased. The ability of MSCs migration obviously decreased when MSCs were treated with U73122, AMD3100. It is concluded that the SDF-1/CXCR4-mediated MSC migration may be related to mitogen-activated protein kinase (MAPK), phosphatidylinositol phospholipase C(PI-PLC) and protein kinase (PKC) signal pathways.
